Microscope Overview

The Carl Zeiss LSM 710 NLO is a multiphoton and confocal point-scanning microscope able to generate high-resolution three-dimensional images of thick specimens with high sensitivity and low photodamage. It is based on Axio-Examiner Z1 upright fixed stage microscope offering a large workspace that can be extended through the easy removal of the condenser, the transmitted NDD lightpath and their carriers.

Applications include, for example, imaging of fixed thick samples, intravital imaging, timelapse imaging, photobleaching experiments, colocalization studies, spectral detection and 3D imaging.

The Zeiss LSM 710 NLO is equipped with tunable Spectra Physics Mai Tai HP DeepSee laser (690-1040 nm) in addition to 6 visible spectrum laser lines (405, 458, 488, 514, 561, 633nm). The multiphoton/two-photon lasers allow for deeper imaging of thick tissue or whole animals.

The LSM 710 scanning unit has a spectral recycling loop, and a QUASAR detection unit (34 PMTs). The QUASAR multichannel photomultiplier detection technology is based on a filter-free system that guides the desired wavelength range to the target detector using adjustable optical wedges and slider light stops. The QUASAR detector allows spectral scanning of fluorescence and emission “fingerprinting” as well as linear unmixing of signals from fluorophores with overlapping emission profiles. Additionally, the system is equipped with four NDD (non-descanned detectors) detectors: 2 GaAsp in reflection and 2 PMT in transmission mode.

The microscope also includes a special Clearing Plan-Neofluar 20x/1.0 objective with 6.4 mm working distance specifically for imaging transparent specimens with nd=1.53
